Cytotoxicity Evaluation of Hydrogel Sheet Dressings Fabricated by Gamma Irradiation: Extract and Semi-Direct Contact Tests

Cytotoxicity presents one of the required criteria in biological evaluation medical devices. In this study, semi-direct contact test was used to evaluate potential cytotoxicity hydrogel wound dressings compared conventional extract test. Three types sheets were fabricated from poly(vinyl alcohol) (PVA) by gamma irradiation: Bare sheets, silver (Ag)-coated and Aloe vera (AV)-coated sheets. test, L929 cells cultured derived elution samples. For situ with samples placed inside transwell inserts above cell monolayers. At endpoint both tests, MTT assay performed, viability determined absorbance formazan. Only bare AV-coated showed 70 % threshold that ensured non-cytotoxicity tests. Ag-coated less than occurred when Ag coating ³ 0.2 mg/cm2. Interestingly, formazan-depleted area underneath sample could be clearly observed The release form nanoparticles confirmed UV-Vis absorption at 420 nm. conclusion, can serve as a reliable alternative evaluating dressings.
